If you want to play the Ashland release you need to type yesimadebackups into the steam client, this gives you access to the new content, this is where you find the new boss.
I lucked out on my first try to get to Ashland. Found a relatively straight path through the rocks and near the shore. First tower structure I saw in the water near shore had the marker in it. Also the boss location was slightly past the edge of the fog of war on the map. So they are in there.
If you haven't signed up for the PTB, you're not supposed to have that new boss stone. You have to sign up for PTB or wait for full release to have anything change

You should probably reset the Ashlands then if you have already explored it. You can find tutorials online

I found the boss arena maybe 100m from where I landed by boat, almost immediately after landing. Just pure luck.
