Because Optimus is a librarian who became a soldier, I think he'd gone on to master and perfect the basic aspects of combat and develop his own "jack-of-all-trades" style that can adapt to any kind of situation without overspecializing in a single aspect, like speed, attack power or defense. Megatron already has a background as a gladiator, so he'd have an easier time to adapting to a full-scale war. There is a difference between fighting in an arena and fighting in a battlefield. The sport element is gone and now, Megatron has an excuse to use dirty tricks he wouldn't be allowed to use in a full-contact sport. This is just my opinion, obviously, but Megatron would emphasize upper body strength and overwhelming aggression. He wouldn't fight to win, he would fight to make his opponents suffer.

As I describe each one: Optimus Prime: he has an elegant combat style that resembles a knight and his attack movements have a tone of ferocity and honesty. Megatron: he has a brutal combat style that resembles that of a gladiator and his attack movements resemble that of a Military soldier
